Structure inside the cell; atomic pathology Speaker: James ‘Jim’ Naismith, University of Oxford, UK Chair: Maria Flocco, AstraZeneca, UK Co-Chair: Stephen McLaughlin, MRC Laboratory of Molecular Biology, UK James ‘Jim’ Naismith states the goal of the Rosalind Franklin Institute to be able to image protein inside tissue. Towards this he gives an overview of the different techniques across a range of resolutions. He highlights the importance of artificial intelligence not only for de-novo protein prediction from sequence but also in recognizing patterns of proteins in cells from cryo-tomography imaging. The describes the workflow for cryo-tomography, emphasizing new methods in labella formation, damage detection and florescence correlation to locate the area of interest. Lastly, he discusses recent work using these methods on the TIR-SAVED which form filaments inside cells induced by tri-cyclic nucleotides.
