Imaging real-time single-molecule dynamics in genome regulation Speaker: Beat Fierz, Ecole Polytechnique Fédérale de Lausanne, Switzerland Chair: David Rueda, Imperial College, London/MRC-LMS, UK Co-Chair: Christopher Batters, MRC Laboratory of Molecular Biology, UK Beat Fierz describes this work to understand the molecular details of how chromatin organisation effects gene regulation. He focuses on the modification of histones by set of enzymes (the ubiquitin ligase PRC1; the PRC methyltransferase; and a deubiquitinase) involved in polycomb gene silencing. Using single-molecule techniques, he examines the effects of ubiquitination on the organization of chromatin using FRET and details the kinetics of binding and the ubiquitation ligation reaction by PRC1 by TIRF.
